Saccharin and Sucralose Protect the Glomerular Microvasculature In Vitro against VEGF-Induced Permeability

چکیده

Diabetic kidney disease (DKD) has become a global health concern, with about 40% of people living type 1 and 2 diabetes mellitus developing DKD. Upregulation vascular endothelial growth factor (VEGF) in the is significant pathology DKD associated increased glomerular permeability. To date, however, current anti-VEGF therapies have demonstrated limited success treating Recent studies shown that artificial sweeteners exhibit potential. The aim this study was therefore to assess effects aspartame, saccharin, sucralose on VEGF-induced leak using an vitro model endothelium. Saccharin but not aspartame protected against Whilst had no effect traditional VEGF signalling, GC-MS analysis sweetener able enter cell exert protective effect. Chemical molecular inhibition sweetener-mediated protection endothelium dependent sweet taste receptor, T1R3. These demonstrate potential for permeability maintain healthy protect glomerulus settings

Vegfa Protects the Glomerular Microvasculature in Diabetes

Vascular endothelial growth factor A (VEGFA) expression is increased in glomeruli in the context of diabetes. Here, we tested the hypothesis that this upregulation of VEGFA protects the glomerular microvasculature in diabetes and that therefore inhibition of VEGFA will accelerate nephropathy. To determine the role of glomerular Vegfa in the development and progression of diabetic nephropathy, w...

Delivery of antioxidant enzyme genes to protect against ischemia/reperfusion-induced injury to retinal microvasculature.

PURPOSE Retinal ischemia/reperfusion (I/R) injury results in the generation of reactive oxygen species (ROS). The aim of this study was to investigate whether delivery of the manganese superoxide dismutase gene (SOD2) or the catalase gene (CAT) could rescue the retinal vascular damage induced by I/R in mice. METHODS I/R injury to the retina was induced in mice by elevating intraocular pressur...

Glomerular Permeability

Ferritin was used as a tracer to investigate glomerular permeability in the nephrotic rat. The results were compared with those previously obtained in normal animals. A nephrotic syndrome was induced by 9 daily injections of the aminonucleoside of puromycin. Ferritin was administered intravenously on the 10th day, and kidney tissue was fixed at intervals of 5 minutes to 44 hours after injection...

Glomerular Permeability

Wistar/Furth rats were made nephrotic by daily administration of amino-nucleoside of puromycin, and the ultrastructural localization of horseradish peroxidase (mol wt 40,000) in the renal glomerulus was studied from 1 min to 20 hr after intravenous injection of the tracer. In control rats, peroxidase permeated the endothelial fenestrae, the basement membrane, and the epithelial slits, and was p...

Cardiac myocyte-specific transgenic ecSOD targets mitochondria to protect against Ca2+ induced permeability transition

ecSOD function has prototypically been associated with the extracellular space due to its secretion and localization to the extracellular matrix. A myocyte-specific ecSOD transgenic mouse has shown that it can also be localized to the myocyte intracellular compartment and is capable of attenuating Reactive oxygen species (ROS) formation and increasing NO bioavailability after ischemia reperfusi...
